Monitoring of Inter-Dose Intervals for Long-Acting Injectable Antipsychotics: A Proposed Protocol for the MIDILIA Trial

چکیده
Aims Service users taking long-acting injectable antipsychotics (LIAs) may experience recurrence of symptoms as they approach trough levels within a steady-state cycle. Limited research exists around symptom variation between peak-to-trough plasma concentrations LIA inter-dose intervals. Different LIAs have variable rates change in dopamine receptor occupancy during this due to differing elimination half-lifes. It is unclear what rate D 2 blockade tolerated by patients at present, which trial aims determine through observing severity differences variation. Methods A real-world observational longitudinal cohort study proposed. Inclusion criteria would be working-age adults (18–65 years) who received five consecutive and timely administrations consistent drug dose. The exclude anyone with significant hepatic or renal impairment, on concurrent oral antipsychotic medication deemed not yet their medication. Serum assays for level will obtained both peak an Expected timings determined derived tmax values from existing pharmacokinetic literature individual drugs. Trough taken 24 hours the next administration being due. Plasma then used calculate expected striatal using EC 50 maximal drugs PET scan data. Symptom assessed completing Positive Negative Scores (PANSS) questionnaires service time LIA. difference these scores plotted alongside concentrations. Results We hypothesize that correlate exponential manner, shorter half-life greater trough. expect able tolerate such degree without emergence symptoms; threshold most can tolerate, assist guiding how effectively reduce discontinue medications. Conclusion This outlines protocol monitor response intervals medication, ultimately aid reducing discontinuing antipsychotics.

Long-acting injectable antipsychotics: recommendations for clinicians.
A major source of limitation to the real effectiveness of antipsychotics is the high rate of patient nonadherence or, more frequently, partial adherence. Using long-acting injectable (LAI) formulations is likely to reduce the impact of such adherence problems. Conversely, the use of LAIs in Canada remains low relative to many other jurisdictions. Based on effectiveness data from randomized cont...

deployable scissor type structures are composed of the so-called scissor-like elements (sles), which are connected to each other at an intermediate point through a pivotal connection and allow them to be folded into a compact bundle for storage or transport. several sles are connected to each other in order to form units with regular polygonal plan views. the sides and radii of the polygons are...
Effectiveness of long-acting injectable antipsychotics: a clinical perspective.
INTRODUCTION The personal and societal costs of schizophrenia spectrum disorders are immense. Affected individuals may experience positive, negative and mood symptoms; medical and substance use comorbidities; and cognitive impairment that significantly impair social and occupational functioning. Globally, schizophrenia is a leading cause of years lost to disability, with a particularly large bu...

Depot formulations are not widely used in everyday practice. This study aimed to assess psychiatrists' attitudes toward the use of long-acting injectable (LAI) antipsychotics in schizophrenia. We interviewed 113 French psychiatrists about the factors that influenced their prescription of LAI antipsychotics. Multidimensional and cluster analyses were used to detect correlations. The most importa...
متن کاملThe Use of Long-Acting Injectable Antipsychotics in Schizophrenia
Long-acting injectable antipsychotics (LAIs) are among the most effective treatments in psychiatry, yet they remain underutilized in clinical practice. Although LAIs are typically used to maintain treatment adherence in patients with chronic schizophrenia, recent research has suggested that they may also provide an effective treatment strategy for patients with early-phase or first-episode dise...
